What Is HPLC Purity Testing? A Guide for Peptide Researchers

High-performance liquid chromatography, or HPLC, is the analytical method most widely used to verify the purity of synthesized peptides. It works by separating the components of a sample as it passes through a column, allowing each compound to be measured individually rather than as part of a mixed solution.

 

During testing, a small sample is dissolved and pushed through the column under pressure. Different molecules travel at different speeds depending on their chemical properties, so the target peptide separates from any leftover reagents, byproducts, or degradation products from synthesis. A detector then records each separated component as a peak, and the area under the main peak relative to all other peaks gives the purity percentage.

 

Purity data matters in a research setting because impurities can behave unpredictably in an experimental system, making results harder to reproduce. A peptide reported at 98% purity contains measurably more background material than one at 99.5%, and that difference can influence sensitive assays.
